I have a lot of experience in doing this (for VW and OS, but the
issues are the same).
I'm on a Mac, and I use the following:
-- For my daily screencasts, I use IShowU to capture what I want. I
restrict the capture window to an 800x600 area, and then use QuickTime
Pro to push out the stuff that gets posted
-- For presentations at events, I've started using ScreenFlow. It
captures the entire screen, but allows for some nice effects like
Picture in Picture, where you can bring in other media (like, say the
video from a camera)
Highlighting sections of the screen with nice transitions
... and other stuff that I haven't really explored
For Windows, I hear good things about Camtasia, but I can't say
anything about it from personal experience. I used Wink for awhile,
and it's ok, but it's all still image aggregation - kind of a "live
slide show" effect
